New insight into cell development and cancer

Dec. 27, 2012 ? Long-standing research efforts have been focused on understanding how stem cells, cells capable of transforming into any type of cell in the body, are capable of being programmed down a defined path to contribute to the development of a specific organ like a heart, lung, or kidney. Research from the University of North Carolina at Chapel Hill School of Medicine has shed new light on how epigenetic signals may function together to determine the ultimate fate of a stem cell.

The study, published December 27, 2012 by the journal Molecular Cell, implicates a unique class of proteins called polycomb-like proteins, or PCL's, as bridging molecules between the "on" and "off" state of a gene. While all of these specialized types of cells share the same genetic information encoded in our DNA, it is becoming increasingly clear that information outside the genome, referred to as epigenetics, plays a central role in orchestrating the reprogramming of a stem cell down a defined path.

Although it is understood that epigenetics is responsible for turning genes "on" and "off" at defined times during cellular development, the precise mechanisms controlling this delicate process are less well understood.

"This finding has important implications for both stem cell biology and cancer development, as the same regulatory circuits controlled by PCL's in stem cells are often misregulated in tumors," said Dr. Greg Wang, senior author of the study and Assistant Professor of Biochemistry and Biophysics in the UNC School of Medicine and a member of UNC Lineberger Comprehensive Cancer Center.

The study, led by postdoctoral research fellows Drs. Ling Cai and Rui Lu in the Wang lab, and Dr. Scott Rothbart, a Lineberger postdoctoral fellow in the lab of Dr. Brian Strahl, Associate Professor of Biochemistry and Biophysics in the UNC School of Medicine and a member of UNC Lineberger Comprehensive Cancer Center, identified that PCL's interact with an epigenetic signal associated with genes that are turned on to recruit a group of proteins called the PRC2 complex which then turn genes off.

"In stem cells, the PRC2 complex turns genes off that would otherwise promote reprogramming into specialized cells of organs like the heart or lungs," said Wang.

In addition to its fundamental role in cellular development, elevated levels of PRC2 have been found in cancers of the prostate, breast, lung, and blood, and pharmaceutical companies are already developing drugs to target PRC2. Wang and colleagues determined that the same mechanisms controlling PRC2 function in stem cells also applies in human cancers.

"The identification of a specific PCL in controlling PRC2 in cancer cells suggests we may be able to develop drugs targeting this PCL to regulate PRC2 function in a more controlled manner that may maintain PRC2 function in stem cells while inhibiting it in the tumor," said Wang.

This research was funded by the National Institutes of Health grants (GM085394 and GM068088), the Department of Defense, the V Foundation for Cancer Research, and the University Cancer Research Fund, and was performed in collaboration with scientists at the University of California at Riverside, Rockefeller University, Memorial Sloan-Kettering Cancer Center, and the Albert Einstein College of Medicine. Study co-authors from UNC also included Bowen Xu, a student in the Wang Lab, and Ashutosh Tripathy, a Research Professor in the Department of Biochemistry and Biophysics.

Wis. police investigate slaying of on-duty officer

WAUWATOSA, Wis. (AP) ? Wisconsin authorities are looking into the death of a suburban Milwaukee police officer who was killed on duty.

Wauwatosa Lt. Gerald Witkowski says dispatchers unsuccessfully tried to reach 30-year-old Jennifer Lynn Sebena for a call about 3 a.m. Monday. He says officers started looking for her and found her dead with several gunshot wounds about 5 a.m. He didn't know if she inside or outside of her car.

Witkowski says the case is considered a homicide. Officers have no suspects.

Witkowski says Sebena enrolled in the police academy in January 2011 and began working solo in July. He says she was married and described her as a great person and officer.

Police are urging witnesses to come forward.

The state Justice Department's criminal investigation unit is helping in the investigation.

Your Email Marketing Is Terrible?. Do Something About It

Companies still don?t understand how email marketing works, and how to best use it. Lets this blog post serve as a quick guide to identifying the most common misconceptions and pitfalls that business need to overcome for a successful email marketing strategy that drives new business and brings old business coming back.

The two types of email marketing ? broadcasts and autoresponders

Broadcasts are when you send a message to an entire list or segment. This is the most common way business use email lists. It is typically used to send coupons, monthly newsletters, or ?deals of the month?. ?Using broadcasts is still better than nothing at all because it keeps your brand top of mind for potential clients and customers, but it is not as powerful as autoresponders.

Autoresponders send a series of predetermined emails at predetermined times to everyone who opt?s in to your email list individually. It is extremely powerful because it can be used to setup a predictable series of engagements after someone takes an initial action used to build value with a client. Value means the ability to sell at a higher price. Most people don?t use autoresponders even close to their true potential because they don?t have the copywriting expertise to get people to open their emails or get them to read the emails and take action.

I know internet marketers that setup autoresponders with over 50 unique emails over the course of months to sell different products and services. The big benefit of a strategy like this is you tend to get predictable results because your prospects are taking the same document sales funnel every single time as opposed to having to worry about whether your sales team can close the deal or not. ?Because of this, your autoresponder can be improved over time to increase open rates, click rates, and track their impact on generating sales. Once you have a system that works, it?s all about getting qualified prospects to do something that nobody finds threatening?..

How to build your list

When you don?t have anyone on an email list it is difficult to spend your time crafting high value content for emails when they are only going to be read by 1 or 5 people. Using email marketing to drive your business means you have to get people to opt-in to getting your information. This requires more than just having a small slot on your website that says ?Join our newsletter?.? This type of call to action has one of the lowest conversion on the web.? You have make getting people to opt-in to your list a primary conversion. ?This is difficult for most people outside of the infoproduct world, because they don?t want less leads, and they think that asking people for only their email instead of a sale or a complete lead form means they miss their chance.

The solution to this problem is to segment your market into different buying stages in your campaigns. For the stages towards the end of the cycle, make you lead forms or checkout button the main call to action that you are trying to get them to take. For people in extremely early stages of the buying cycle, consider using an email opt-in with an incentive as the main call to action. Here are several examples where email opt-in would work well as the main call to action:

Most likely they will also need to purchase a badger hair shave brush as well as shave soap. These can all be done in a consultative manner as long as value is provided along the way.

Tip: A great way to get the ball rolling and turn searchers like this into paying customers is to drop in an entry level purchase that is extremely cheap and available only to those that have opted in. The goal is not to devalue your brand by discounting, but by lowering the barrier of entry for a person to conduct business with you for the first time. Once they do, they will be more likely to do it again.

Here is another example of how this strategy works in the B2B arena.

This would typically not be the type of search that has the intent of obtaining a consultation from a sales person from an inventory tracking software vendor. By providing a suite of free tools or white papers that are helpful to them at their stage in the buying cycle, you can obtain their email and nurture the lead until it becomes ready to engage in a consultation.

How effective is this strategy?

It works incredible well, but it takes time and a lot of effort to put together. When you use copywriting to do your selling for you, your sales funnel will only be as good as the copy used in it. Attempting to use an opt-in strategy to drive your business when your sales copy and content is mediocre is the same as expecting a bad sales person to drive the growth of your company. A good content funnel requires significant time and effort to put together, but it is an investment that pays off.

What are the steps that need to take place to start working towards an effective funnel?

Think of the content funnel for your email marketing strategy as a bridge that your customers must cross. If one spot in the bridge is missing some planks, nobody gets across. You have to systematically remove the barriers that are stopping people from cross your bridge.? You can do that by focusing on specific steps of the funnel.

Step 1: Develop a landing page for a specific audience and content topic.

Your goal will be to get the opt-in rate as high as you possibly can by split testing different content and layouts on the page. ?Remember, the more people you get to opt in to you funnel, the more of a chance your content will have to succeed.? Most people would find it hard to spend $1000 on writing up a single email content piece to send to 10 people, but if you are sending it to 20,000 it may not be a big deal.? Having a larger list creates a virtuous cycle of value.

Step 2: Track you email open and click rates and split test them to get them to improve.

Email is about delivery, open rates, and click rates. Go one step at a time and keep working your numbers up.? Most people make the mistake of using fancy email templates. Most people open their emails on their phone these days and all the major email programs remove images by default. Save them the trouble and use plain text split up in twitter like bite size pieces that make it easy to read the whole thing. Step 3: Moving them forward towards the sale

If you don?t know the path that customers will take to get to a sale then don?t expect them to. Every step of your process should have clear next step.? Many companies who can get people to listen to them and open their emails because of great content still fail to turn that interest into sales. Don?t be afraid to ask for the sale. Step 4: Closing the deal

This part tends to be the easiest. Anyone that gets this far into your funnel wants to buy from you. You just have to make sure not to screw it up.? Just make sure not to distract with another goal or action until they have completed the one that you asked them to take by coming here. NEWTOWN, Conn. (AP) ? The family of Noah Pozner was mourning the 6-year-old, killed in the Newtown school massacre, when outrage compounded their sorrow.

Someone they didn't know was soliciting donations in Noah's memory, claiming that they'd send any cards, packages and money collected to his parents and siblings. An official-looking website had been set up, with Noah's name as the address, even including petitions on gun control.

Noah's uncle, Alexis Haller, called on law enforcement authorities to seek out "these despicable people."

"These scammers," he said, "are stealing from the families of victims of this horrible tragedy."

It's a problem as familiar as it is disturbing. Tragedy strikes ? be it a natural disaster, a gunman's rampage or a terrorist attack ? and scam artists move in.

It happened after 9/11. It happened after Columbine. It happened after Hurricane Katrina. And after this summer's movie theater shooting in Aurora, Colo.

Sometimes fraud takes the form of bogus charities asking for donations that never get sent to victims. Natural disasters bring another dimension: Scammers try to get government relief money they're not eligible for.

"It's abominable," said Ken Berger, president and CEO of Charity Navigator, which evaluates the performance of charities. "It's just the lowest kind of thievery."

Noah Pozner's relatives found out about one bogus solicitation when a friend received an email asking for money for the family. Poorly punctuated, it gave details about Noah, his funeral and his family. It directed people to send donations to an address in the Bronx, one that the Pozners had never heard of.

It listed a New York City phone number to text with questions about how to donate. When a reporter texted that number Wednesday, a reply came advising the donation go to the United Way.

The Pozner family had the noahpozner.com website transferred to its ownership. Victoria Haller, Noah's aunt, emailed the person who had originally registered the name. The person, who went by the name Jason Martin, wrote back that he'd meant "to somehow honor Noah and help promote a safer gun culture. I had no ill intentions I assure you."

Alexis Haller said the experience "should serve as a warning signal to other victims' families. We urge people to watch out for these frauds on social media sites."

Consumer groups, state attorneys general and law enforcement authorities call for caution about unsolicited requests for donations, by phone or email. They tell people to be wary of callers who don't want to answer questions about their organization, who won't take "no" for an answer, or who convey what seems to be an unreasonable sense of urgency.

"This is a time of mourning for the people of Newtown and for our entire state," Connecticut Attorney General George Jepsen said in a statement this week. "Unfortunately, it's also a time when bad actors may seek to exploit those coping with this tragedy."

But scam artists know that calamity is fertile ground for profit, watered by the goodwill of strangers who want to help and may not be familiar with the cause or the people they're sending money to.

After the shootings at Columbine High School in Littleton, Colo., scammers asked for credit card donations for victims' families. After the 9/11 attacks, the North American Securities Administrators Association warned investors to be wary of Internet postings encouraging them to invest in supposed anti-terrorist technologies.

In 2006, the FBI warned about an email widely circulated after the Sago, W.Va., mine explosion, which claimed to be from a doctor treating one of the survivors and asking for donations to cover medical bills.

"As was learned after the tragic events of 9/11/01, the tsunami disaster, and more recently with Hurricane Katrina, unscrupulous cyber criminals have shown the desire and means to exploit human emotion by attempting to defraud the public when they are perceived to be most vulnerable," the FBI said at the time.

This fall, the police in Aurora, Colo., accused a local woman of trying to profit off the deadly movie theater rampage by a gunman who killed 12 people. The woman told people that she was the caretaker for a little girl named Kadence, whose mother had died in the shooting. The police said the child was made up. The scam unraveled when a donor got a phone call from what seemed to be a woman imitating a child's voice.

When the government doled out disaster aid after Hurricane Katrina, scammers asked for money to rebuild houses they never lived in or to pay benefits for relatives who never existed.

The government later set up the National Center for Disaster Fraud to try to root out such scams in the federal relief programs administered after Hurricanes Katrina, Rita and Wilma. It has since expanded its mandate to other disasters.

The cases brought since then by the Justice Department sketch a colorful picture of fraud:

? A woman who filed for small-business disaster benefits after the 2010 Gulf Coast oil spill, even though she'd sold the business before the accident.

? A judge and a commissioner in Texas who, after Hurricane Ike, were accused of awarding debris removal contracts to a company in return for kickbacks. The judge also commandeered a 155-kilowatt generator meant for the county to power his convenience store, according to the government.

? A pastor who submitted inflated claims to a government-funded program that reimbursed groups sheltering Hurricane Katrina evacuees.

Bob Webster, spokesman for the NASAA, knows the sad pattern.

"We know cons try to cash in on headlines, and any who would even think about stooping to capitalize on the tragedy in Newtown are the lowest of the low," he said.

NEW YORK (AP) ? Radio talk show host Laura Ingraham will soon be back on the air after taking a short break.

Courtside Entertainment Group says it will distribute "The Laura Ingraham Show" starting Jan. 2. Meanwhile, she will boost her audience through a deal with Lauchpad Digital Media, which will stream the three-hour daily program and make it available for download on podcast.

Talkers magazine says the conservative Ingraham is the most-listened-to woman on radio, with an estimated 5.75 million listeners a week. Her show, launched in 2001, is now heard on more than 300 radio stations nationally.

She is also a Fox News contributor and principal guest host of "The O'Reilly Factor."

Ingraham began a brief hiatus from her show last month after ending a deal with its former distributor.

Climate warming unlikely to cause near-term extinction of ancient Amazon trees, but multiple threats to the forest remain

Dec. 13, 2012 ? A new genetic analysis has revealed that many Amazon tree species are likely to survive human-caused climate warming in the coming century, contrary to previous findings that temperature increases would cause them to die out.

However, the authors of the new study warn that extreme drought and forest fires will impact Amazonia as temperatures rise, and the over-exploitation of the region's resources continues to be a major threat to its future. Conservation policy for the Amazon should remain focused on reducing global greenhouse-gas emissions and preventing deforestation, they said.

The study by University of Michigan evolutionary biologist Christopher Dick and his colleagues demonstrates the surprising age of some Amazonian tree species -- more than 8 million years -- and thereby shows that they have survived previous periods as warm as many of the global warming scenarios forecast for the year 2100.

The paper is scheduled for online publication Dec. 13 in the journal Ecology and Evolution. The new study is at odds with earlier papers, based on ecological niche-modeling scenarios, which predicted tree species extinctions in response to relatively small increases in global average air temperatures.

"Our paper provides evidence that common Amazon tree species endured climates warmer than the present, implying that -- in the absence of other major environmental changes -- they could tolerate near-term future warming under climate change," said Dick, an associate professor of ecology and evolutionary biology and acting director of the U-M Herbarium.

But study co-author Simon Lewis of University College London and the University of Leeds cautioned that "the past cannot be compared directly with the future."

"While tree species seem likely to tolerate higher air temperatures than today, the Amazon forest is being converted for agriculture and mining, and what remains is being degraded by logging and increasingly fragmented by fields and roads," Lewis said. "Species will not move as freely in today's Amazon as they did in previous warm periods, when there was no human influence. Similarly, today's climate change is extremely fast, making comparisons with the past difficult.

"With a clearer understanding of the relative risks to the Amazon forest, we conclude that direct human impacts, such as forest clearance for agriculture or mining, should remain a focus of conservation policy," Lewis said. "We also need more aggressive action to reduce greenhouse gas emissions in order to minimize the risk of drought and fire impacts to secure the future of most Amazon tree species."

Dick and his colleagues used a molecular clock approach to determine the ages of 12 widespread Amazon tree species, including the kapok and the balsa. Then they looked at climatic events that have occurred since those tree species emerged. In general, they inferred that the older the age of the tree species, the warmer the climate it has previously survived.

The researchers determined that nine of the tree species have been around for at least 2.6 million years, seven have been present for at least 5.6 million years, and three have existed in the Amazon for more than 8 million years.

"These are surprisingly old ages," Dick said. "Previous studies have suggested that a majority of Amazon tree species may have originated during the Quaternary Period, from 2.6 million years ago to the present."

Air temperatures across Amazonia in the early Pliocene Epoch (3.6 million to 5 million years ago) were similar to Intergovernmental Panel on Climate Change projections for the region in 2100 using moderate carbon-emission scenarios. Air temperatures in the late Miocene Epoch (5.3 to 11.5 million years ago) were about the same as IPCC projections for the region in 2100 using the highest carbon-emission scenarios.

The 12 tree species used in the study are broadly representative of the Amazon tree flora. Primary forest collection sites were in central Panama, western Ecuador and Amazonian Ecuador. Additional collections were made in Brazil, Peru, French Guiana and Bolivia. Other plant samples were obtained from herbarium specimens.

"The most lasting finding of our study may be the discovery of ancient geographic variation within widespread species, indicating that many rain forest tree species were widely distributed before the major uplift of the northern Andes," said co-author Eldredge Bermingham of the Smithsonian Tropical Research Institute.

To determine the age of each tree species, the researchers extracted and sequenced DNA from plant samples, then looked at the number of genetic mutations contained in those sequences. Using a molecular clock approach and population genetic models, they estimated how long it would take for each of the tree populations to accumulate the observed number of mutations, which provided a minimum age for each species.

"An important caveat is that because we've been in a cold period over the past 2 million years -- basically the whole Quaternary Period -- some of the trees' adaptations to warmth tolerance may have been lost," Dick said. "Additional research is needed to test whether this has occurred."

The Ecology and Evolution paper is titled "Neogene origins and implied warmth tolerance of Amazon tree species." In addition to Dick, Bermingham and Lewis, Mark Maslin of University College London is a co-author. Financial support for the research was provided by the Smithsonian Tropical Research Institute, the University of Michigan, the National Science Foundation and the Royal Society.

Poll: Even science doubters say world is warming

WASHINGTON (AP) ? Nearly 4 out of 5 Americans now think temperatures are rising and that global warming will be a serious problem for the United States if nothing is done about it, a new Associated Press-GfK poll finds.

Belief and worry about climate change are inching up among Americans in general, but concern is growing faster among people who don't often trust scientists on the environment. In follow-up interviews, some of those doubters said they believe their own eyes as they've watched thermometers rise, New York City subway tunnels flood, polar ice melt and Midwestern farm fields dry up.

Overall, 78 percent of those surveyed said they thought temperatures were rising and 80 percent called it a serious problem. That's up slightly from 2009, when 75 percent thought global warming was occurring and just 73 percent thought it was a serious problem. In general, U.S. belief in global warming, according to AP-GfK and other polls, has fluctuated over the years but has stayed between about 70 and 85 percent.

The biggest change in the polling is among people who trust scientists only a little or not at all. About 1 in 3 of the people surveyed fell into that category.

Within that highly skeptical group, 61 percent now say temperatures have been rising over the past 100 years. That's a substantial increase from 2009, when the AP-GfK poll found that only 47 percent of those with little or no trust in scientists believed the world was getting warmer.

This is an important development because, often in the past, opinion about climate change doesn't move much in core groups ? like those who deny it exists and those who firmly believe it's an alarming problem, said Jon Krosnick, a Stanford University social psychologist and pollster. Krosnick, who consulted with The Associated Press on the poll questions, said the changes the poll shows aren't in the hard-core "anti-warming" deniers, but in the next group, who had serious doubts.

"They don't believe what the scientists say, they believe what the thermometers say," Krosnick said. "Events are helping these people see what scientists thought they had been seeing all along."

Phil Adams, a retired freelance photographer from Washington, N.C., said he was "fairly cynical" about scientists and their theories. But he believes very much in climate change because of what he's seen with his own eyes.

"Having lived for 67 years, we consistently see more and more changes based upon the fact that the weather is warmer," he said. "The seasons are more severe. The climate is definitely getting warmer."

"Storms seem to be more severe," he added. Nearly half, 49 percent, of those surveyed called global warming not just serious but "very serious," up from 42 percent in 2009. More than half, 57 percent, of those surveyed thought the U.S. government should do a great deal or quite a bit about global warming, up from 52 percent three years earlier.

But only 45 percent of those surveyed think President Barack Obama will take major action to fight climate change in his second term, slightly more than the 41 percent who don't think he will act.

Overall, the 78 percent who think temperatures are rising is not the highest percentage of Americans who have believed in climate change, according to AP polling. In 2006, less than a year after Hurricane Katrina devastated New Orleans, 85 percent thought temperatures were rising. The lowest point in the past 15 years for belief in warming was in December 2009, after some snowy winters and in the middle of an uproar about climate scientists' emails that later independent investigations found showed no manipulation of data.

Broken down by political party, 83 percent of Democrats and 70 percent of Republicans say the world is getting warmer. And 77 percent of independents say temperatures are rising. Among scientists who write about the issue in peer-reviewed literature, the belief in global warming is about 97 percent, according to a 2010 scientific study.

The AP-GfK poll was conducted Nov. 29-Dec. 3 by GfK Roper Public Affairs and Corporate Communications. It involved landline and cellphone interviews with 1,002 adults nationwide. Results for the full sample have a margin of error of plus or minus 3.9 percentage points; the margin of error is larger for subgroups.

The latest AP-GfK poll jibes with other surveys and more in-depth research on global warming, said Anthony Leiserowitz, director of Yale University's Project on Climate Change Communication. He took no part in the poll.

When climate change belief was at its lowest, concerns about the economy were heightened and the country had gone through some incredible snowstorms and that may have chipped away at some belief in global warming, Leiserowitz said. Now the economy is better and the weather is warmer and worse in ways that seem easier to connect to climate change, he said.

"One extreme event after another after another," Leiserowitz said. "People have noticed. ... They're connecting the dots between climate change and this long bout of extreme weather themselves."

Thomas Coffey, 77, of Houston, said you can't help but notice it.

"We use to have mild temperatures in the fall going into winter months. Now, we have summer temperatures going into winter," Coffey said. "The whole Earth is getting warmer and when it gets warmer, the ice cap is going to melt and the ocean is going to rise."

He also said that's what he thinks is causing recent extreme weather.

"That's why you see New York and New Jersey," he said, referring to Superstorm Sandy and its devastation in late October. "When you have a flood like that, flooding tunnels like that. And look at how long the tunnel has been there."
